Shenise Johnson scored 19 points, grabbed 15 rebounds and had 5 assists for the Royal Comets.
This is the 17th double-double of the year for Johnson. The only game in which she did not achieve a double-double was against Penfield when she fouled out in the fourth quarter after the referee gave her a technical foul.
Osha Jones and LaDonya Rolle combined for 12 rebounds for Rush-Henrietta (15-3).
Penfield is 15-3.
"Our bread and butter is just good man-to-man defense. We've still got a long way to go to repeat as Section V champs", Rush-Henrietta head coach Steve Shepanski said.
Rush-Henrietta have been Section V champs the past four years, three out of the four championship games having been won against Penfield.
The Royal Comets lost nine players, four of their five starters, from last year's team due to graduation.
In a matchup earlier this year Penfield defeated Rush-Henrietta 50-49 in overtime after Yohannes' 3-pointer in the final nine seconds of play.
Rush-Henrietta and Penfield tied for sixth place in the Democrat and Chronicle large schools poll.
Tonight was senior night for host Rush-Henrietta.
